摘要:
研究从 Candida Mogii细胞中,提取木糖还原酶 ( XR)的实验条件及 XR的催化动力学 .( 1)当破碎停留时间为 3min、离心转速为 12 0 0 0 r· min-1时,提取液中 XR的酶活保留较好 .( 2 )适当的 K+能够提高酶活,而 Mg2 +对 XR有抑制作用 .( 3)所得粗提液中的 XR具备一定的催化能力,可满足实际应用的要求 .( 4 )所得粗提液中的 XR半衰期约为 50 h,其稳定性还不足以实现长期连续催化生产 .
Abstract:
Candida Mogii cells are taken as object, from which xylose reductase(XR) is extracted. A study is made on experimental condition of extraction and catalytic kinetics of XR. As indicated by the study: (1) XR in crude extract keeps a fairly good enzyme activity under a crush retaining for 3 min. and a centrifugal speed up to 1 200 r·min -1; (2) enzyme activity of XR can be enhanced by K + in suitable concentration, but it will be inhibited by Mg 2+; (3) XR in crude extract has fixed catalytic force for satisfying the demand of practical use; and (4) XR in crude extract shows a half life about 50 hrs, its stability is inadequate for realizing long time continuous catalytic production.
