洁霉素产生菌不论生长在含有甘氨酸的S培养基或不含甘氨酸的S培养基的菌丝中,其细胞壁均可被溶菌酶溶解而释放出原生质体。甘氨酸浓度为1.0%破壁效果最好,溶菌酶的作用浓度以2.5mg/ml为最佳。原生质体在Hb培养基上能够很好地再生。用P培养基配制的40%(w/v)PEG1000溶液,能有效地诱导洁霉素产生菌原生质体融合。融合重组频率达6.7%。
